Familial Intracranial Aneurysm in Newfoundland: Clinical and Genetic Analysis

Objective:
Intracranial aneurysm (IA) is an expansion of the weakened arterial wall that is often asymptomatic until rupture, resulting in subarachnoid hemorrhage. Here we describe the high prevalence of familial IA in a cohort of Newfoundland ancestry. We began to investigate the genetic etiology of IA in affected family members, as the inheritance of this disease is poorly understood.
Methods:Whole exome sequencing was completed for a cohort of 12 affected individuals from two multiplex families with a strong family history of IA. A filtering strategy was implemented to identify rare, shared variants. Filtered variants were prioritized based on validation by Sanger sequencing and segregation within the families.
Results:In family R1352, six variants passed filtering; while in family R1256, 68 variants remained, so further filtering was pursued. Following validation by Sanger sequencing, top candidates were investigated in a set of population controls, namely, C4orf6 c.A1G (p.M1V) and SPDYE4c.C103T (p.P35S). Neither was detected in 100 Newfoundland control samples.
Conclusion:Rare and potentially deleterious variants were identified in both families, though incomplete segregation was identified for all filtered variants. Alternate methods of variant prioritization and broader considerations regarding the interplay of genetic and environmental factors are necessary in future studies of this disease.

Background
A range of endophenotypes characterise psychosis, however there has been limited work understanding if and how they are inter-related.
MethodsThis multi-centre study includes 8754 participants: 2212 people with a psychotic disorder, 1487 unaffected relatives of probands, and 5055 healthy controls. We investigated cognition [digit span (N = 3127), block design (N = 5491), and the Rey Auditory Verbal Learning Test (N = 3543)], electrophysiology [P300 amplitude and latency (N = 1102)], and neuroanatomy [lateral ventricular volume (N = 1721)]. We used linear regression to assess the interrelationships between endophenotypes.
ResultsThe P300 amplitude and latency were not associated (regression coef. −0.06, 95% CI −0.12 to 0.01, p = 0.060), and P300 amplitude was positively associated with block design (coef. 0.19, 95% CI 0.10–0.28, p < 0.001). There was no evidence of associations between lateral ventricular volume and the other measures (all p > 0.38). All the cognitive endophenotypes were associated with each other in the expected directions (all p < 0.001). Lastly, the relationships between pairs of endophenotypes were consistent in all three participant groups, differing for some of the cognitive pairings only in the strengths of the relationships.
ConclusionsThe P300 amplitude and latency are independent endophenotypes; the former indexing spatial visualisation and working memory, and the latter is hypothesised to index basic processing speed. Individuals with psychotic illnesses, their unaffected relatives, and healthy controls all show similar patterns of associations between endophenotypes, endorsing the theory of a continuum of psychosis liability across the population.

Two cyclones for sampling dilute aerosols in the field are described. Their advantages are their robustness and reliability, the high degree of concentration achieved, and the ease with which they will provide a long uninterrupted sequence of samples.
Methods of correcting estimates of cloud concentration for transfer lag in the cyclone are derived.
The cyclones were constructed by J. Nott. We are indebted to G. J. Harper and E. J. Morris for allowing us to make use of their observations, and to Dr J. S. Paterson (Superintendent, Allington Farm) for providing the realistic aerosol at our convenience.

In a previous paper (Twort, Baker, Finn & Powell, 1940) we were able to show that the bactericidal properties of phenolic -aerosols were correlated with their physical characters, though our experiments were not sufficiently numerous or varied to enable us to estimate separately the effect of the different factors operative. Here we propose to describe similar tests which we have carried out on the germicidal smokes used by Twort & Baker (1940, 1941), namely, those produced by smouldering cardboard impregnated with potassium nitrate, and by dropping incense and Peru balsam on to hot metal surfaces. There were also used mixtures obtained by distilling Peru balsam under reduced pressure: fraction I, b. p. 109–110° C; fraction II, b. p. 120–138° C; fraction III, bp 148–155° C, at 3 mm. Hg, and the residue from this operation.

The evaporation rates of small particles in aerosols of the following substances have been measured: 4−n−hexyl-1:3-resorcinol, 1, 1, 1-trichloro-2, 2−di (p−chlorphenyl)ethane (‘DDT’), 1, 1, 1−trichloro-2-p−chlorphenyl-2-o−chlorphenyl-ethane (‘op′-DDT’), γ-hexachloro-cyclo-hexane (‘BHC’, ‘Lindane’, ‘Gammexane’), ‘Dieldrin’ and azobenzene.
The slit ultra-microscope method was used; a consideration of the experimental conditions leads to the conclusion that the results are of reliable quantitative significance.

The principle of the aerosol centrifuge is described, and an elementary mathematicaltreatment is given.
The relations between the behaviour of the centrifuge and its dimensions, speed and airflow have been tested experimentally on three machines. It is shown that the mathe-matical formulae given are an adequate basis for design.
Various applications are mentioned.

A form of ultramicroscope apparatus is described which enables the evaporation of mist droplets to be followed for long periods. A rough method of estimating the size distribution in aerosols is also given.
The rates of evaporation of particles of various phenol solutions have been studied, the solutions chosen being of possible interest as aerial germicides. It is shown that the typical evaporation curve for a binary mixture consists of three parts, which in actual cases need not all be present. Sometimes the more detailed history of an evaporation can be deduced from the results.
The approximate effective size distributions in mists of hexyl-resorcinol—propylene-glycol solutions have been measured and compared with the evaporation rates.
It is shown that the efficiency of aerial germicides is partly a function of their volatility, and three rough classes are distinguished. But biological evidence is adduced which suggests that the solubility of the germicides in water may be of equal importance, and that the liquid in which the germicide is dissolved is not without effect.
A centrifuge is described which enables the maximum particle size in a mist to be limited to a desired value.
Methods of estimating quantitatively germicidal aerosols are discussed, and a method described which has been found to give reasonably accurate results in practice. Tentative experiments have been made to determine whether the materials studied are likely to cause corrosion of metals, etc., after pro-longed use.
The collision theory of aerial disinfection is discussed, as is also the applicability of Langmuir's treatment of the evaporation of fine particles in mixtures.
The development of the experimental technique involved in the biological investigations appertaining to air disinfection has been discussed, together with the numerous fallacies arising and the means by which these have been overcome.
The construction of a special pair of experimental chambers is described, and details given, both as regards the bacteria and germicides, of the exact experimental conditions under which the most consistent results can be obtained.
The requisite characteristics of germicidal mixtures which could be utilized for air disinfection are considered, and examples of suitable germicides and solvents are given. The method of recording and evaluating the results of a varied assortment of experiments is explained to facilitate inter se comparison.
Examples are given of the lethal effectiveness in the air of several germicides in broth emulsions, both with and without the presence of serum, on stock cultures of selected types of bacteria. The sensitivity to the germicides of the bacteria making up the flora of the average normal saliva is compared with that of broth emulsions of bacteria.
10% hexyl-resorcinol dissolved in propylene-glycol+0·05 % sulphonated lorol (“S2”) was the most effective all round germicidal mixture of those tested. Various other germicides were found equal in germicidal activity in the air, but for reasons stated have been deemed unsuited for use in the presence of man.
The relationship between germicidal efficiency in the air and mist particle size and persistence has been studied by means of centrifuged mists. Particles of 0·5–1·0μ radius are shown to be the most useful in dealing with bacterial particles containing organic matter. Laboratory cultures, emulsified in broth, have been sterilized in mist dispersions by droplets of bactericides at least as small as 0·25μ radius.
Tests on the penetrating abilities of bactericidal mists and bacteria through certain cloths have been performed.
Experiments to demonstrate the surface sterilizing properties of aerosols have been performed, and examples of differences in effect on horizontal and vertical surfaces are noted.
The degree of tolerance of man and animals (mice) for a number of phenolic substances and some organic solvents has been compared with the degree of tolerance of bacteria for the same substances.
The tests on man have been confined to the detection by the special senses of the substances suspended in the air as a fine mist.
The tests on animals were designed to show acute and chronic effects, the results being recorded in terms of clinical symptoms and pathological changes in the organs. A record was also kept of the findings as regards protozoal parasites of the host, and compared with the normal incidence among ordinary laboratory stock.
Tests on bacteria were instituted for the purpose of gaining information on the host-parasite tolerance ratio for our germicides when in the air and when in the test-tube.
Effects of primary tillage and soil amendment practices on pearl millet yield and nutrient uptake in the Sahel of West Africa

The main objective of this study was to determine the best soil amendment and tillage practices for sustainable millet yield and grain and stover quality. The treatments included tillage practices (immediate-, late- and no-till) and soil amendments (sheep manure plus urine, manure, millet stover (stalks, leaf blades and leaf sheaths) and millet stover ash) in factorial combinations with fertilizer nitrogen levels of 0, 15 and 30 kg ha−1 plus controls. Results showed that (i) higher yields were obtained in tilled plots than in no-till plots; (ii) tillage timing may not be a significant yield determining factor; (iii) the application of animal urine resulted in significantly higher yield and greater nitrogen, phosphorus and potassium uptake than the application either of manure alone or of millet stover. Urine application (ruminant urine contains virtually no phosphorus), which elevates soil pH especially during the first week after application, may have resulted in the dissolution of phosphorus from the aluminium–iron complexes of kaolinitic clays. This is corroborated by the significantly higher phosphorus uptake from the manure-plus-urine plots than from plots amended with either manure alone or millet stover. Long-term implications of ‘mining’ soil phosphorus with repeated applications of animal urine in these fragile ecosystems remains unclear.
Amorphous Silicon Image Sensor Arrays

We have developed a technology for 2D matrix-addressed image sensors using amorphous silicon photodiodes and thin film transistors. We have built a small prototype, having 192×192 pixels with a 20μm pixel pitch, and assessed its performance. The nip photodiodes can have dark current densities of less than 1011 A.cm-2 (up to 5V reverse bias) and peak quantum efficiencies of 88% (at 580nm). We operated the sensor in real time mode at high speed (50 Hz frame rate and 64μS line time). The image sensor has a low noise performance giving a dynamic range in excess of 104. The maximum crosstalk is about 2%, which allows at least 50 grey levels. The bottom contact of the photodiode acts as a light shield from light through the substrate, which enables the sensor to be operated as an intimate contact image sensor to image a document placed directly on top of the array. In this mode, the CTF was 75% at 2 lp.mm1. Good quality images are demonstrated in both front projection and intimate contact imaging modes.
